Kursets målsætning
The aim of the course is to provide a practical as well as a theoretical introduction to basic biotechnology for plant breeding. Based on acquired basic skills, students are introduced to further principles of planning, management, and report writing for biotech projects.
 
Kursusindhold
A general introduction to major techniques and principles of plant breeding biotechnology is given during the first semester, comprising:
* Micropropagation.
Germplasm establishment in vitro, multiplication techniques, in vitro storage, and re-establishment in soil.
* Callus and cell suspensions.
Cell culture establishment, maintenance and plant regeneration.
* Haploid production.
Anther/microspore culture, chromosome elimination, and ovule/ovary culture.
* Protoplast technology.
Protoplast isolation, culture, fusion, and regeneration of plants.
* Genetic transformation.
Agrobacterium mediated transformation, direct gene uptake including biolistic systems.
* Molecular markers.
RFLP, RAPDS, AFLPs, and microsatellites.
Map construction, QTL-mapping, and positional cloning.
 
Undervisningsform
The course is comprised of three parts: A basic part, supervised exercises, and a project. During the basic part, which lasts one month, the students fundamental skills are established. This enables them to perform independent design and preparation of culture media, sterilisation and dissection of explants, as well as other laboratory operations, based on descriptions from the literature. During the two months of supervised exercises, students conduct experiments within the major topics: micropropagation, cell culture, haploids, protoplast and transformation, and molecular markers. Supervised exercises use plant material prepared in advance and prepared structural protocols to be completed by the students. Part of the first semester is spent planning the course project based on the students ideas and literature searches. The second semester is devoted to the experimental project with plant material and follows the plans prepared by the students. A final written report based on the course project is used for the final exam.
